A comparative study between different methods to assess cell viability
Since the introduction of the trypan blue exclusion method of determining cell viability, there have been many technological advances to improve the reliability and reduce manual steps.

Manual cell counting
Advantage:
- Low cost of equipment.
Disadvantages:
- Low precision & accuracy.
- Labor-intensive & dye-based.
Coulter counter
Advantage:
- Label-free.
Disadvantages:
- Unreliable viability determination.
- Data interpretation is difficult.
Flow cytometer
Advantage:
- Viability determination.
Disadvantages:
- Dye-based.
- Difficult to differentiate between apoptotic and live cells.
Automated counting
Advantage:
- Good precision & accuracy.
Disadvantages:
- Dye-based & sample intensive.
- Out of focus images result in lost data.
“On-line” counting
Advantage:
- Automated sampling.
Disadvantages:
- Space-intensive.
- Consumes a lot of sample.
“In-line” counting
Advantage:
- Continuous data and no waste.
Disadvantages:
- Viability determination is difficult.
- Cell adherence to probe over time.
